One-Step Suicide Substrate Inactivation Kinetics of a Ping-Pong Reaction with One Substrate Undergoing Disproportionation: A Theoretical Approach with Approximate Solutions

چکیده

Understanding the kinetic mechanism of enzyme inactivation by suicide substrate is relevance for optimal design new drugs with pharmacological and therapeutic applications. Suicide usually occurs via a two-step mechanism, although there are enzymes such as peroxidase catalase in which H2O2 happens single step. The approximate solution ordinary differential equation (ODE) system one step kinetics uni–uni reaction following irreversible Michaelis–Menten model was previously analytically solved when accumulation substrate–enzyme complex negligible, however not more models, ping-pong reaction, present two active states during catalytic turnover. To solve this issue, theoretical approach followed, standard quasi-steady state reactant stationary approximations were invoked. These allowed solving ODE undergoing disproportionation also present. Although analytical solutions rather unwieldy, they still valuable qualitative analyses to explore time course products identify that irreversibly reacted reaction.

The kinetics of succinyl coenzyme A synthetase from Escherichia coli. A reaction with a covalent enzyme-substrate intermediate not exhibiting "ping-pong" kinetics.

Succinyl coenzyme A synthetase from Escherichia coli does not exhibit a steady state kinetic pattern indicative of “ping-pong” kinetics, despite the fact that catalytic participation of a phosphorylated enzyme covalent intermediate has been established. Instead, kineti.c patterns are consistent with the sequential addition of all substrates, to form a quaternary complex, before the release of a...

Derivation of rate equations for multisite ping-pong mechanisms with ping-pong reactions at one or more sites.

It has become clear as the result of the pioneering studies of Northrop on transcarboxylase (l), and the later studies in Lardy’s (2) and Utter’s (3) laboratories on pyruvate carboxylase that biotin-containing enzymes have two-site ping-pong mechanisms in which biotin is carboxylated at one site, and in turn carboxylates a substrate at the second site. The rate equations for such mechanisms hav...
